A série, baseada nos livros de Sara Shepard, conta a história de um grupo de amigas que morrem de inveja de Alison, a garota mais popular da escola, e sonham em tomar seu lugar. Aria (Lucy Hale), Hanna (Ashley Benson), Spencer (Troian Bellisario) e Emily (Shay Mitchell) guardam um segredo que só Alison conhece, porém ela desaparece e as garotas não ficam tristes com isso.

Depois de três anos, quando as garotas seguiram com suas vidas, começam a receber várias mensagens ameaçadoras, prometendo revelar o segredo que elas guardam. Todas as mensagens vêm assinadas com a letra “A”, o que deixará as garotas desesperadas.
